ACTIVATED PRODUCING HOCL NEUTROPHILS REVEALED BY FLOW CYTOMETRY AND CONFOCAL MICROSCOPY WITH CELESTINE BLUE B

Abstract (English):
HOCl obrazuetsya pri stimulyacii neytrofilov blagodarya aktivnosti mieloperoksidazy. Dlya selektivnoy spektrofotometricheskoy detekcii HOCl predlozhen celestinovyy siniy B (CB). Cel'yu dannoy raboty yavilas' razrabotka fluorescentnyh metodov ocenki produkcii HOCl neytrofilami. Produkt reakcii CB s HOCl fluoresciroval pri 578 nm (vozbuzhdenie 487 nm). Metodom konfokal'noy mikroskopii poluchili izobrazheniya aktivirovannyh neytrofilov, okrashennyh CB. Pri protochnoy citometrii fluorescenciya aktivirovannyh 50 nM PMA neytrofilov byla v 5 raz vyshe (p < 0,05), chem u kletok bez aktivatora.
